采用RACE方法从鹤望兰(Strelitzia reginae Banks)黄色花萼中克隆到1个LCYB基因(lycopene beta-cyclase,Sr LCYB)。其c DNA全长为1 908 bp(Gen Bank收录号:KT428725),具有完整的开放阅读框(open reading frame,ORF),共1 515个碱基,编码504个氨基酸。鹤望兰Sr LCYB的氨基酸序列与其他物种都有一定的同源性,其中与百合的LCYB同源性最高,达到82%。系统进化树分析显示,鹤望兰Sr LCYB与芭蕉蛋白亲缘关系较近。应用荧光定量PCR分析表明,Sr LCYB在黄色花萼中高表达,且在花发育的始花期和盛花期表达量最高。应用UPLC(ultra performance liquid chromatography)对β-胡萝卜素含量进行分析,类β-胡萝卜素在黄色花萼中含量最高,且在花发育的盛花期含量最高。结果表明,Sr LCYB基因表达与β-胡萝卜素含量成正相关。
One LCYB gene (lcL LCYB) was cloned from the yellow calyx of Strelitzia reginae Banks by RACE method. The full-length cDNA of cDNA was 1 908 bp (Gen Bank Accession No. KT428725) with a complete open reading frame (ORF) of 1 515 bases encoding 504 amino acids. The amino acid sequence of Streptomyces przewalskii Sr LCYB shared some homology with other species. The homology of LCYB to that of lily was 82%. Phylogenetic tree analysis showed that the relationship between Strelitzia reginae Sr LCYB and banana protein was close. The quantitative PCR analysis showed that Sr LCYB was highly expressed in the yellow calyx and the highest expression was observed at the flowering and flowering stages. The content of β-carotene was analyzed by ultra performance liquid chromatography (UPLC). The content of β-carotene was highest in the yellow calyx and highest in the flowering stage. The results showed that Sr LCYB gene expression and β-carotene content was positively correlated.
