目的:建立同时测定升阳散火汤中葛根素、阿魏酸、异阿魏酸、蛇床子素和异欧前胡素5种活性成分含量的RP-HPLC法。方法:采用Diamonsil C18(200mm×4.6mm,5μm)色谱柱,流动相为甲醇-4%醋酸水溶液,梯度洗脱;流速1.0mL.min-1;紫外检测波长310nm;柱温35℃。结果:葛根素、阿魏酸、异阿魏酸、蛇床子素和异欧前胡素线性范围分别为15.6~156μg·mL-1(r=0.9995),0.660~6.60μg·mL-1(r=0.9997),8.96~89.6μg·mL-1(r=0.9995),6.30~63.0μg·mL-1(r=0.9993),1.32~13.2μg·mL-1(r=0.9996);方法平均回收率(n=9)分别为99.7%,100.6%,100.2%,100.6%,99.8%。结论:本法简便、准确,重复性好,可为升阳散火汤质量评价提供依据。
OBJECTIVE: To establish an RP-HPLC method for simultaneous determination of five active ingredients, Puerarin, Ferulic acid, Isoferulic acid, Osthol and Isoimperatorin in Shengyang Sanhuo Decoction. Methods: Diamonsil C18 (200mm × 4.6mm, 5μm) was used as the mobile phase. The mobile phase consisted of methanol-4% acetic acid aqueous solution with gradient elution. The flow rate was 1.0mL.min-1. The UV detection wavelength was 310nm and the column temperature was 35 ℃. Results: The linear ranges of puerarin, ferulic acid, isoferulic acid, osthole and isoimperatorin were 15.6 ~ 156μg · mL -1 (r = 0.9995) and 0.660 ~ 6.60μg · mL -1 (r = 0.9997), 8.96 ~ 89.6μg · mL -1 (r = 0.9995), 6.30 ~ 63.0μg · mL -1 (r = 0.9993) and 1.32 ~ 13.2μg · mL -1 (n = 9) were 99.7%, 100.6%, 100.2%, 100.6% and 99.8% respectively. Conclusion: This method is simple, accurate and reproducible, which can provide the basis for the quality evaluation of Shengyang Powder.
