目的通过点眼和结膜下注射贝伐单抗两种不同的给药途径,抑制兔角膜碱烧伤后的新生血管,探讨该药的这两种给药方式的疗效。方法普通白色家兔50只,取2只为正常对照组,其余48只右眼碱烧伤后随机分为三组,每组16只:A组为碱烧伤对照组,B组为点眼组,用贝伐单抗(5mg/ml)点眼,3次/d,C组为结膜下注射组,用贝伐单抗(2.5mg/0.1ml)结膜下注射。在不同的时间点(第1、4、7、14d)测量角膜新生血管的生长情况及免疫组化检测角膜血管内皮生长因子(vascular endothelial growth factor,VEGF)蛋白的表达。结果 B、C组的CNV相对于A组得到明显抑制,CNV的长度和面积及VEGF蛋白的表达差异有统计学意义,而B组和C组之间没有明显差异。结论两种用药方法对于抑制角膜新生血管没有明显差异。
OBJECTIVE: To observe the effects of the two modes of administration by observing two different routes of administration of bevacizumab by conjunctival injection and subconjunctival injection to inhibit neovascularization after corneal alkaline burn in rabbits. Methods Fifty normal white rabbits were selected as normal control group and the remaining 48 eyes were randomly divided into three groups (n = 16 in each group): alkali burn group A, control group B, Bevacizumab (5 mg / ml) was given to the eyes for 3 times / d. Group C was subconjunctival injected with subconjunctival injection of bevacizumab (2.5 mg / 0.1 ml). Corneal neovascularization was measured at different time points (days 1, 4, 7 and 14), and immunohistochemistry was used to detect the expression of corneal endothelial growth factor (VEGF). Results The CNV in groups B and C was significantly inhibited compared with that in group A, while the length and area of CNV and the expression of VEGF protein were significantly different. There was no significant difference between group B and group C. Conclusion There is no significant difference between the two methods in inhibiting corneal neovascularization.
