目的研究空腹血糖受损(IFG)患者和Ⅱ型糖尿病(T2DM)患者肠道菌群整体结构多样性,筛选出可能与T2DM相关的细菌。方法利用末端限制性片段长度多态性(T-RFLP)技术检测正常人群、IFG患者和T2DM患者肠道菌群整体结构;运用多元统计方法偏最小二乘判别分析(PLS-DA)对T-RFLP数据进行分析,筛选出与T2DM可能相关的细菌。结果 T2DM组肠道菌群整体结构的个体差异较对照组和IFG组增大,初步筛选出3种可能与Ⅱ型糖尿病相关的细菌,Bacillus sp.ES-SL-2、Fusobacterium nucleatum和Thermosyntropha sp.L-60。结论本研究初步筛选出了可能与Ⅱ型糖尿病相关的肠道菌株,证实T-RFLP联合PLS-DA应用于Ⅱ型糖尿病早期发现和疾病状态监控的可行性。
Objective To study the overall structure diversity of intestinal microflora in patients with impaired fasting glucose (IFG) and type 2 diabetes mellitus (T2DM), and to screen for possible bacteria associated with T2DM. Methods T-RFLP was used to detect the intestinal flora in normal population, IFG patients and T2DM patients. Multivariate statistical method was used to analyze the effect of PLS-DA on T- RFLP data analysis, screening and T2DM may be related to bacteria. Results The individual differences in the overall structure of intestinal microflora in T2DM group were higher than those in control group and IFG group. Three kinds of bacteria which may be associated with type Ⅱ diabetes, Bacillus sp. ES-SL-2, Fusobacterium nucleatum and Thermosyntropha sp were screened out. L-60. CONCLUSIONS: This study initially screened intestinal strains that may be associated with type II diabetes and confirmed the feasibility of using T-RFLP in combination with PLS-DA in early detection of type II diabetes and monitoring of disease status.
