支气管扩张药茶碱的血浆浓度超过治疗范围可引起致命的全身性惊厥发作。发生这种神经毒性表现的血药浓度的个体差异很大。这可能是因为一些病人存在茶碱致惊毒性的某些敏感因素,如肾衰就是这种危险因素。鉴于发热或环境引起的发热均可提高对惊厥的敏感性,本文用大鼠酵母致热模型,观察了发热对茶碱神经毒性的影响。动物为雄性Lewis大鼠,180±6g。实验前分笼单个喂养6天,室温23~25℃,12h光照与12h黑暗交替进行。实验前2天,行颈静脉硅胶导管插管。实验前12h,颈背部皮下注射15%Brewer′s酵母10ml/kg,以产生发热,对照组注射生理盐水。用直肠温度计测量深部体温。注射酵母可升高体温1.2±0.4℃。
