目的:研究五味降压方对自发性高血压大鼠(SHR)的降压作用,并对五味降压方起效的血中暴露成分进行研究。方法:五味降压方2.6 g.kg-1大鼠灌胃给药(相当于人临床日用量的12倍),每日1次,连续4周,每周测定大鼠血压1次;末次测定血压后,腹主动脉采血,分离血浆,反相高效液相色谱法建立五味降压方以及血中多成分指纹图谱,探明降压疗效反应呈现下的暴露成分。结果:五味降压方从第2周起表现出明显的降血压作用(P<0.01);对血中多成分指纹图谱进行分析,发现五味降压方在降压疗效发生时,有紫外吸收成分进入血液。结论:五味降压方药效反应明显,血中可见具有紫外吸收的暴露成分,其暴露成分的确切性质有待于通过化学研究工作进一步阐明。
Objective: To study the antihypertensive effect of Wuwei Jiangya Decoction on spontaneous hypertensive rats (SHR) and to study the blood exposure components of Wuwei Jiangya antihypertensive prescription. Methods: The rats of 2.6 g.kg-1 of Wuwei Jiangya Antihypertensive Formula were intragastrically administrated (equivalent to 12 times of daily clinical dosage) once daily for 4 weeks and the blood pressure of rats was measured once a week. The last test Blood pressure, the abdominal aorta blood collection, plasma separation, reverse phase high performance liquid chromatography established five anti-hypertensive side and blood multi-component fingerprinting to identify the antihypertensive response to the presence of exposed components. Results: Wuwei Hypotensive Prescription showed obvious antihypertensive effect from the second week (P <0.01). Analyzing the multi-component fingerprints in blood, it was found that Wuwei Hypotensive Prescription had antihypertensive effect, Into the blood. Conclusion: Wuwei Jiao Jiao Prescription has obvious pharmacodynamic response, visible in the blood with UV absorption of exposed components, the exact nature of exposure components to be further clarified through the chemical research work.
