目的:探讨虎金颗粒对免疫性肝纤维化大鼠肝组织中Ⅰ、Ⅲ型胶原沉积和基质金属蛋白酶抑制剂-1(TIMP-1)表达的影响。方法:建立猪血清诱导的大鼠免疫性肝纤维化模型,并给予虎金颗粒进行治疗,采用免疫组化法检测肝组织中Ⅰ、Ⅲ型胶原蛋白的表达,并通过逆转录聚合酶链反应(RT-PCR)半定量法测定肝组织中TIMP-1mRNA的表达。结果:虎金颗粒可使受试动物肝组织Ⅰ、Ⅲ型胶原增生减少,纤维间隔变窄,肝组织TIMP-1 mRNA表达降低,与模型组比较差异显著(P<0.01)。结论:虎金颗粒可通过抑制肝纤维化大鼠肝脏Ⅰ、Ⅲ型胶原蛋白和TIMP-1的合成表达,而发挥抗纤维化作用。
Objective: To investigate the effects of Hujin Granule on the expression of collagen type Ⅰ and Ⅲ and the expression of tissue inhibitor of metalloproteinase-1 (TIMP-1) in the liver of immunological hepatic fibrosis rats. Methods: A rat model of autoimmune liver fibrosis induced by porcine serum was established and treated with Hujin granule. Immunohistochemistry was used to detect the expression of type Ⅰ and type Ⅲ collagen in liver tissues. The expression of collagen type Ⅰ and Ⅲ was detected by reverse transcription polymerase chain reaction (RT-PCR) semi-quantitative determination of TIMP-1 mRNA expression in liver tissue. Results: Hujin granule could reduce the proliferation of type Ⅰ and type Ⅲ collagen, narrow the fiber space and decrease the expression of TIMP-1 mRNA in liver tissue of experimental animals. The difference was significant (P <0.01). Conclusion: Hujin Granule can exert its anti-fibrosis effect by inhibiting the synthesis of type Ⅰ, Ⅲ collagen and TIMP-1 in hepatic fibrosis rats.
