目的观察体外培养传代的毛乳头细胞表达干细胞因子(stem cell factor,SCF)情况;并观察SCF对体外培养的毛乳头细胞生长曲线的影响。方法应用免疫组化及流式细胞技术测量不同代次的毛乳头细胞SCF的表达变化情况,采用MTT比色法观察SCF对毛乳头细胞生长增殖的影响。结果低传代培养的毛乳头细胞SCF的表达较强,随着传代次数的增多,表达逐渐减弱。SCF组与对照组相比,细胞生长曲线的停滞期减少,提前进入对数生长期,并且OD值与对照组相比明显升高,差异有统计学意义(P<0.05)。结论 SCF可以促进体外培养的人毛乳头细胞的增殖。
Objective To observe the expression of stem cell factor (SCF) in cultured dermal papilla cells in vitro and observe the effect of SCF on the growth curve of dermal papilla cells cultured in vitro. Methods The expression of SCF in dermal papilla cells of different generations was measured by immunohistochemistry and flow cytometry. The effects of SCF on the proliferation and proliferation of dermal papilla cells were observed by MTT assay. Results The expression of SCF in dermal papilla cells of low subculture was stronger than that of control. The expression of SCF was decreased with the passage of times. Compared with the control group, the cell growth arrest phase decreased in SCF group and entered the logarithmic growth phase in advance, and the OD value was significantly higher than that in the control group (P <0.05). Conclusion SCF can promote the proliferation of human dermal papilla cells cultured in vitro.
