目的 探讨人巨细胞病毒 (HCMV)对人骨髓粒 -单系祖细胞 (GM -CFUc)集落形成能力的影响。方法 采用造血祖细胞培养技术 ,以HCMVAD169持续攻击骨髓GM -CFUc ,观察计数GM -CFUc集落、簇的产率及集落的大小和维持的时间。结果 ①HCMV感染组GM -CFUc集落和簇的产率明显低于对照组 (P <0 0 1) ;②HCMV感染GM -CFUc后 ,大集落的产率明显低于对照组 ;③集落维持的时间缩短 ,而集落出现峰值的时间无明显差异。结论 在体外HCMV对GM -CFUc生长、集落的形成有明显抑制作用 ,提示HCMV感染患儿粒细胞减少可能与HCMV损伤GM -CFUc有关
Objective To investigate the effect of human cytomegalovirus (HCMV) on the colony formation ability of human bone marrow granulocyte-monocyte progenitor cells (GM-CFUc). Methods The hematopoietic progenitor cell culture technique was used to continuously challenge the bone marrow GM-CFUc with HCMVAD169 to observe the yield of colonies and clusters, the size of colonies and the maintenance time of GM-CFUc colonies. Results ① The yield of GM-CFUc colonies and clusters in HCMV infection group was significantly lower than that in control group (P <0.01); ② After GM-CFUc infection with HCMV, the yield of large colony was significantly lower than that of control group; ③The time of colony maintenance was shortened , While there was no significant difference in the peak time of the colonies. Conclusions HCMV could inhibit the growth of GM-CFUc and the formation of colonies in vitro, suggesting that the neutropenia in children with HCMV infection may be related to the impairment of GM-CFUc by HCMV
