单克隆抗体是近年发展起来的一种新技术。本实验应用酶标记抗体法,鉴定抗hCG单克隆抗体的特异性,并与抗hCG多克隆抗体比较,以积累使用单克隆抗体的经验。 取人早期妊娠(40~60天)胎盘绒毛、大鼠和小鼠垂体各4例,固定于4%多聚甲醛,4℃下孵育24小时。经磷酸缓冲液浸洗3天后,脱水、透明、石蜡包埋。另人垂体4例是我校病理教研室提供的、经10%福尔马林固定后石蜡包埋的组织块。全部标本均作厚5μ的连续切片,同时进行免疫组织化学染色。用作第一抗体的抗hCG多克隆抗体是我校第一医院制备
