目的 :探讨 5 -氟尿嘧啶 (5 - Fu)对大肠癌细胞 Fas蛋白表达水平的调控作用。方法 :采用流式细胞术检测了培养的 9株细胞 (6株为人大肠癌细胞 ) Fas蛋白的表达情况 ,并观察了 5 - Fu(2 6 0 mol/ L )处理后大肠癌细胞 Fas蛋白表达水平的变化。结果 :在检测的 6株人大肠癌细胞中 ,有 5株肿瘤细胞表面表达 Fas蛋白 ;用临床浓度的 5 - Fu处理后 ,HCT- 116和 Wi Dr的Fas蛋白的表达水平显著上调。 结论 :5 - Fu可上调肿瘤细胞 Fas表达水平 ,由此可能增加其对 Fas介导的机体免疫监视作用的敏感性。
Objective: To investigate the regulation of Fas protein expression in colorectal cancer cells by 5-fluorouracil (5-Fu). Methods: The expression of Fas protein in 9 cultured cells (6 colorectal cancer cells) was detected by flow cytometry. The expression of Fas protein in colorectal cancer cells treated with 5-Fu (260mol/L) was also observed. The level of change. RESULTS: Fas protein was expressed on the surface of 5 tumor cells in 6 human colorectal cancer cells. After treatment with clinical concentrations of 5-FU, the expression of Fas protein in HCT-116 and Wi Dr was significantly up-regulated. Conclusion : 5-Fu can up-regulate the expression of Fas in tumor cells, which may increase its sensitivity to Fas-mediated immune surveillance.
