目的通过下调脑挫伤细胞中laminin,观察其对新生大鼠皮质神经干细胞迁移能力的影响。方法设计合成下调大鼠laminin的siRNA并转染至大鼠脑挫伤细胞,realtime PCR检测脑挫伤细胞中laminin基因水平siRNA干扰效率;westernblot检测脑挫伤细胞中laminin蛋白水平siRNA干扰效率;迁移实验检测神经干细胞迁移率。结果大鼠脑挫伤细胞转染下调laminin的siRNA后,laminin基因表达水平下降(71.35±4.26)%,laminin蛋白水平较对照组下降(50.1±3.45)%;迁移实验显示siRNA组神经干细胞迁移能力下降,P<0.05。结论下调脑挫伤细胞中laminin表达,可以抑制神经干细胞迁移能力,因此尽量保存脑外伤中挫伤组织有利于损伤区域神经再生。
Objective To investigate the effect of laminin on the migration of cortical neural stem cells in neonatal rats by down-regulating laminin in brain contusion cells. Methods siRNA targeting laminin was synthesized and transfected into rat brain contusion cells. Real-time PCR was used to detect siRNA interference efficiency of laminin gene in brain contusion cells. Western blot was used to detect siRNA interference efficiency of laminin protein in brain contusion cells. Stem cell migration. Results The expression of laminin decreased (71.35 ± 4.26)% and the laminin protein level decreased (50.1 ± 3.45)% in brain contusion cells transfected with siRNA of laminin. Migration assay showed that the migration ability of NSCs in siRNA group decreased , P <0.05. Conclusions The down-regulation of laminin expression in brain contusion cells can inhibit the migration of neural stem cells. Therefore, it is beneficial to preserve the contusion tissues of traumatic brain injury in the regeneration of injured area.
