目的:探讨溃疡性结肠炎(UC)患者外周血T细胞亚群及γδ-T细胞的表达在UC发病中的作用。方法:用流式细胞仪双染法检测13例正常人、10例活动性溃疡性结肠炎(AUC)及12例非活动性溃疡性结肠炎(IAUC)患者外周血及PHA刺激后的T细胞亚群及γδ-T细胞的表达。结果:PHA刺激后,CD4+细胞在AUC及IAUC组均较对照组显著增加(P均<0.05);AUC及IAUC组间差异无显著性,但AUC组的CD_4 ̄+Leu8 ̄-(辅助细胞)较PHA刺激前显著增加(P<0.05);γδ-T细胞(即TCRδl ̄+CD_3 ̄+)在AUC组较对照组及IAUC组均显著减少(P均<0.05),而-T细胞的部分抗原表达(即TCRδ1 ̄+CD_3 ̄+)在AUC组及IAUC组均较对照组显著减少(P分别<0.01,<0.05),AUC及IAUC组间差异无显著性;外周血TCRδ1 ̄+CD_4 ̄+占总TCRδ1 ̄+细胞百分数在AUC组较对照组及IAUC组均显著增加(P均<0.05),PHA刺激后该细胞在AUC及IAUC组较对照组显著增加(P均<0.05)·结论:UC患者γδ-T细胞的增殖性降低,可能反映了γδ-T细胞对外来抗原免疫监视作用?
Objective: To investigate the role of peripheral blood T cell subsets and γδ-T cells in the pathogenesis of UC in patients with ulcerative colitis (UC). Methods: Flow cytometry was used to detect peripheral blood of 13 normal subjects, 10 patients with active ulcerative colitis (AUC) and 12 patients with inactive ulcerative colitis (IAUC) and T cells stimulated by PHA Subpopulations and γδ-T cells. Results: After stimulated with PHA, the percentage of CD4 + cells in AUC and IAUC groups increased significantly (P <0.05), but there was no significant difference between AUC and IAUC groups. However, CD_4 + Leu8- (P <0.05); γδ-T cells (TCRδl ~ + CD_3 ~ +) in AUC group were significantly decreased compared with control group and IAUC group (all P <0.05) The expression of T antigen (TCRδ1 ~ + CD_3 ~ +) in AUC group and IAUC group was significantly lower than that in control group (P <0.01, <0.05, respectively) Peripheral blood TCRδ1 ~ + CD_4 ~ + total TCRδ1 ~ + cell percentage in AUC group than the control group and IAUC group were significantly increased (P all <0.05), PHA stimulation in the AUC and IAUC group compared with the control group increased significantly (P <0.05 respectively) · Conclusion: γδ-T cell proliferation in patients with UC decrease may reflect the foreign antigens, immune surveillance γδ-T cells?
