目的:研究益妇宁软胶囊(YFN)对去卵巢大鼠生殖内分泌-免疫功能的影响。方法:3月龄SD雌性大鼠60只,50只行双侧卵巢切除术,随机分为YFN高剂量组(2.0 g/kg)、中剂量组(1.0 g/kg)、低剂量组(0.5 g/kg)、阳性对照组(己烯雌酚0.1 mg/kg)及模型对照组(model),每组10只,另10只为假手术组(sham)。术后4 w开始给药,给药12 w后用放射免疫法测定血清雌二醇(E2)、卵泡刺激素(FSH)、黄体生成素(LH)、睾酮(Te)、孕激素(P)水平及白介素-2(IL-2)含量,用流式细胞术测定血T淋巴细胞CD亚群。结果:①与假手术组比较,模型组大鼠血清E2、Te、P水平显著下降(P<0.01),FSH、LH水平显著上升(P<0.05或P<0.01);血T淋巴细胞百分含量显著下降(P<0.05),CD4+T淋巴细胞亚群显著减少(P<0.01),CD4+/CD8+比值显著降低(P<0.01),CD8+T淋巴细胞亚群无显著性变化;血清白介素-2含量显著下降(P<0.01)。②益妇宁治疗后去卵巢大鼠血清E2、P水平显著上升(P<0.01),FSH、LH显著下降,血T淋巴细胞亚群CD3+、CD4+和CD4+/CD8+比值均显著升高,血清IL-2水平显著提高,高剂量组较中、低剂量组作用显著。结论:益妇宁软胶囊能升高去卵巢大鼠血清性激素水平,下调FSH、LH水平,改善失衡的T淋巴细胞亚群,促进IL-2的分泌,调节去卵巢大鼠紊乱的生殖内分泌-免疫网络。
Objective: To study the effect of Yifuning soft capsule (YFN) on reproductive endocrine-immune function in ovariectomized rats. METHODS: Sixty SD female rats aged 3 months and 50 patients undergoing bilateral ovariectomy were randomly divided into high-dose YFN (2.0 g/kg), middle-dose (1.0 g/kg), and low-dose groups (0.5 g/kg), positive control group (diethylstilbestrol 0.1 mg/kg) and model control group (model), 10 in each group, and the other 10 were sham. The administration was started 4 w after surgery, and serum estradiol (E2), follicle stimulating hormone (FSH), luteinizing hormone (LH), testosterone (Te), and progesterone (P) were measured by radioimmunoassay after 12 weeks. Levels and interleukin-2 (IL-2) content were determined by flow cytometry. RESULTS: 1Compared with the sham-operated group, serum E2, Te and P levels in the model group decreased significantly (P<0.01), FSH and LH levels increased significantly (P<0.05 or P<0.01); The content decreased significantly (P<0.05), CD4+ T lymphocyte subsets decreased significantly (P<0.01), CD4+/CD8+ ratio decreased significantly (P<0.01), CD8+ T lymphocyte subsets did not change significantly; serum interleukin The content of -2 decreased significantly (P < 0.01). 2 Serum level of E2 and P in ovariectomized rats increased significantly (P<0.01), FSH, and LH decreased significantly after Yifuning treatment. The ratios of CD3+, CD4+, and CD4+/CD8+ in T lymphocyte subsets increased significantly. The level of -2 increased significantly, and the effect of the high dose group was significantly higher than that of the middle and low dose groups. Conclusion: Yifuning soft capsule can increase serum sex hormone levels in ovariectomized rats, down-regulate FSH and LH levels, improve imbalanced T lymphocyte subsets, promote IL-2 secretion, and regulate reproductive endocrine disorders in ovariectomized rats.
