探讨利用噬菌体随机肽库筛选到的AKA相关模拟抗原对RF阴性类风湿关节炎患者的诊断价值。将间接荧光法检测AKA阳性的RA患者混合血清,经33.3%饱和硫酸铵盐析纯化IgG,以此多克隆抗体为配基,对噬菌体递呈的随机12肽库进行淘筛,将筛选到的阳性噬菌体克隆调整至一定浓度包被酶标反应板,通过间接ELISA法检测RA血清标本,与RF检测结果进行比较。结果是筛选到的AKA模拟抗原对RA诊断的特异性为97.4%,敏感性为56.7%;RF阴性患者中AKA阳性率为52.4%。因此,筛选到的AKA模拟抗原对RF阴性的RA患者是一个有价值的补充诊断指标。
To investigate the diagnostic value of AKA-associated mimic antigens screened by phage random peptide library in patients with RF-negative rheumatoid arthritis. The results showed that the specificity of AKA mimic antigen screening for RA was 97.4% and the sensitivity was 56.7%. The positive rate of AKA in RF-negative patients was 52.4%. Therefore, the screened AKA mimic antigen is a valuable complementary diagnostic indicator for RF-negative RA patients.
