目的探讨内毒素/脂多糖(LPS)对小鼠肺、肝、心氧化损伤的影响。方法24只ICR纯系雄性小鼠随机分为5,10,15 mg/kg 3个LPS染毒组和1个对照组,每组6只。采用腹腔注射方法进行LPS染毒,对照组注射生理盐水,测定染毒后肺、肝、心3种脏器中抗氧化物质和脂质过氧化(LPO)的水平。结果LPS腹腔注射引起所有受试脏器谷胱甘肽(GSH)含量下降,LPO水平升高,且随着LPS染毒剂量的增大,各器官组织氧化损伤程度加重。经t-检验,中、高剂量LPS处理组与对照组比较差异均有统计学意义。LPS对不同脏器引起的损伤程度不同,存在器官差异,总体上对肝、肺的损伤较为严重。结论LPS可引起小鼠肺、肝、心等多种脏器氧化损伤,且损伤程度与内毒素剂量成正相关。
Objective To investigate the effects of endotoxin / lipopolysaccharide (LPS) on lung, liver and heart oxidative damage in mice. Methods Twenty four ICR male mice were randomly divided into 3 LPS groups (5, 10, 15 mg / kg) and 1 control group, with 6 mice in each group. The rats in LPS group were injected intraperitoneally with LPS and the control group with normal saline. The levels of antioxidant substances and lipid peroxidation (LPO) in lung, liver and heart were measured after exposure. Results The intraperitoneal injection of LPS caused a decrease of glutathione (GSH) and LPO in all the organs tested, and the oxidative damage of various organs increased with the increase of LPS dose. The t-test, medium and high-dose LPS treatment group compared with the control group differences were statistically significant. LPS on different organs caused by different degrees of damage, there are organ differences, the overall damage to the liver and lungs more serious. Conclusion LPS can cause oxidative damage of lung, liver and heart in mice, and the degree of injury is positively correlated with the dosage of endotoxin.
