目的分析Nud CD1在结直肠癌患者外周血中的表达水平与临床病理特征及预后生存的关联,探讨Nud CD1在结直肠癌早期诊断及病程预后评估中的作用。方法采用实时定量PCR法(q PCR)检测结直肠癌(100例)及结直肠腺瘤患者(58例)、健康志愿者(100例)外周血中Nud CD1 m RNA表达,采用ROC曲线判断Nud CD1对结直肠癌的诊断价值,计算Youden指数最大值确定结直肠癌诊断阈值的Nud CD1 m RNA水平;采用Bivariate Correlation(双变量相关)Pearson检验分析Nud CD1 m RNA表达水平与结直肠癌患者临床病理特征及3年生存期的相关性;采用Kaplan-Meier生存分析(Log-rank检验)分析Nud CD1 m RNA表达与结直肠癌患者3年生存期相关性,绘制生存曲线。结果结直肠癌患者外周血Nud CD1 m RNA水平相对于结直肠腺瘤患者外周血(P<0.05)、健康志愿者外周血(P<0.01)显著高表达;结直肠癌患者外周血Nud CD1 m RNA相对表达量与肿瘤直径、原发灶浸润深度、神经脉管累及、分化程度、淋巴结转移数、远处转移及TNM分期(P均<0.01)呈显著正相关,与3年生存期呈负相关(P<0.01);Kaplan-Meier生存分析(Log-rank检验)显示Nud CD1 m RNA高表达组3年生存时间显著低于低表达组(P<0.01)。结论结直肠癌患者外周血Nud CD1 m RNA表达水平与临床病理特征及3年生存期具有显著关联性,Nud CD1可作为一种有价值的结直肠癌早期筛查及预后评估的标记物。
Objective To analyze the relationship between the expression of Nud CD1 in peripheral blood of patients with colorectal cancer, the clinicopathological features and prognosis of colorectal cancer, and to explore the role of Nud CD1 in the early diagnosis of colorectal cancer and the evaluation of the prognosis. Methods Real-time quantitative PCR (qPCR) was used to detect the expression of Nud CD1 mRNA in colorectal cancer (100 cases) and colorectal adenoma (58 cases) and healthy volunteers (100 cases) CD1 on the diagnosis of colorectal cancer, calculate the maximum Youden index to determine the diagnostic threshold of colorectal cancer Nud CD1 m RNA levels; Bivariate Correlation (Pearson correlation) Pearson test analysis Nud CD1 m RNA expression and clinical colorectal cancer patients Pathological features and 3-year survival. The Kaplan-Meier survival analysis (Log-rank test) was used to analyze the relationship between the expression of Nud CD1 m RNA and the 3-year survival of patients with colorectal cancer, and the survival curves were drawn. Results The level of Nud CD1 m RNA in peripheral blood of patients with colorectal cancer was significantly higher than that of patients with colorectal adenoma (P <0.05), and that of healthy volunteers (P <0.01) The relative expression of RNA was positively correlated with tumor diameter, depth of primary tumor invasion, involvement of vessel, differentiation, lymph node metastasis, distant metastasis and TNM stage (all P <0.01), but negatively correlated with 3-year survival (P <0.01). The Kaplan-Meier survival analysis (Log-rank test) showed that the 3-year survival time of high expression Nud CD1 m RNA group was significantly lower than that of low expression group (P <0.01). Conclusion The expression of Nud CD1 m RNA in peripheral blood of patients with colorectal cancer is significantly correlated with the clinicopathological characteristics and 3-year survival. Nud CD1 can be used as a valuable marker for early screening and prognosis of colorectal cancer.
