目的:探讨过表达肿瘤相关基因LRP16对卵巢癌细胞株HO8910生长的影响。方法:将含有LRP16基因真核表达载体pcDNA3.1-LRP16及空载体质粒转染卵巢上皮癌细胞株HO8910使其稳定过表达,用MTT法测各组细胞生长曲线,用Western blot方法检测E-钙黏着素(E-cadherin)蛋白的表达。结果:LRP16基因过表达对HO8910细胞增殖无影响,LRP16基因过表达的HO8910细胞的E-钙黏着素(E-cadherin)蛋白表达水平明显下降。结论:LRP16基因对卵巢上皮癌细胞HO8910的增殖无影响,但使E-钙黏着素(E-cadherin)表达明显下降。
Objective: To investigate the effect of over-expression of tumor-associated gene LRP16 on the growth of ovarian cancer cell line HO8910. METHODS: Eukaryotic expression vector pcDNA3.1-LRP16 containing LRP16 gene and empty vector plasmid were transfected into ovarian epithelial carcinoma cell line HO8910 to stably overexpress the cells. MTT assay was used to measure the cell growth curve of each group. Western blot was used to detect E- Expression of E-cadherin protein. RESULTS: Overexpression of LRP16 had no effect on the proliferation of HO8910 cells. The expression of E-cadherin in HO8910 cells over-expressing LRP16 gene was significantly decreased. Conclusion: LRP16 gene has no effect on the proliferation of ovarian epithelial carcinoma HO8910 cells, but the expression of E-cadherin is significantly decreased.
