目的:研究糖肝康对糖尿病脂肪肝模型大鼠肝脏组织中AdipoR_2 mRNA表达的影响。方法:高糖+高脂+小剂量链脲佐菌素腹腔注射建立糖尿病脂肪肝大鼠模型,并设对照组进行观察。药物干预12周后,采用RT-PCR方法检测糖肝康对肝脏组织中AdipoR_2mRNA表达的影响。结果:模型大鼠的肝脏组织中AdipoR_2 mRNA表达水平较低(0.143±0.046),糖肝康可上调其在肝脏组织中的mRNA表达(小剂量组为0.202±0.052,大剂量组为0.212±0.049),与模型对照组比较,差异有统计学意义(P<0.05),且大剂量组表达高于降糖甲对照组(0.176±0.065)。结论:糖尿病脂肪肝的糖脂代谢紊乱可能与AdipoR_2的低水平表达相关,糖肝康可通过上调AdipoR_2 mRNA表达水平,在糖、脂代谢等方面到积极作用,值得进一步深入研究。
Objective: To study the effect of TangGankang on the expression of AdipoR_2 mRNA in the liver of diabetic rats with fatty liver. Methods: Diabetic fatty liver rat models were established by intraperitoneal injection of high-glucose + high-fat + low-dose streptozotocin (STZ) and the control group was observed. Twelve weeks after drug intervention, RT-PCR was used to detect the effect of Tanggankang on the expression of AdipoR_2mRNA in liver tissue. Results: The expression of AdipoR_2 mRNA in the liver tissue of model rats was lower (0.143 ± 0.046), and the expression of mRNA in liver tissues was up-regulated by Tanggankang (0.202 ± 0.052 in low-dose group and 0.212 ± 0.049 in high-dose group ), Compared with the model control group, the difference was statistically significant (P <0.05), and high-dose group was higher than the hypoglycemic control group (0.176 ± 0.065). CONCLUSION: The disorder of glycolipid metabolism in diabetic fatty liver may be related to the low level expression of AdipoR_2. Tanggikang may play an active role in up-regulation of AdipoR_2 mRNA expression in glucose and lipid metabolism, which deserves further study.
