目的探讨狼疮肾炎(LN)外周血OX40,OX40L mRNA表达水平及抗C1q抗体水平。方法采集80例系统性红斑狼疮患者及40例正常对照者外周血,采用ELISA法检测抗C1q抗体,采用荧光定量逆转录聚合酶链反应(RT-PCR)测定外周血单个核细胞(PBMC)中OX40,OX40L mRNA表达水平。结果LN患者抗C1q抗体水平为(9.11±2.46)ng/mL,显著高于非LN患者的(3.12±0.97)ng/mL及正常对照者的(1.12±0.33)ng/mL,差异有统计学意义(P<0.05)。LN患者PBMCs OX40,OX40L mRNA水平分别为(8±4)×106拷贝/μL和(4±3)×103拷贝/μL,显著高于非LN患者[(4±3)×103及(2±2)×103]和正常对照者[(4±2)×103及(2±1)×103],差异有统计学意义(P<0.01)。结论外周血OX40,OX40L mRNA表达及抗C1q抗体与LN活动度相关,其在LN的发生、发展中可能起一定作用。
Objective To investigate the expression of OX40 and OX40L mRNA in peripheral blood of lupus nephritis (LN) and the level of anti-C1q antibody. Methods Totally 80 peripheral blood samples from patients with systemic lupus erythematosus (SLE) and 40 normal controls were collected. Anti-C1q antibody was detected by ELISA. Peripheral blood mononuclear cells (PBMCs) were detected by real-time fluorescence quantitative reverse transcription polymerase chain reaction OX40, OX40L mRNA expression levels. Results The level of anti-C1q antibody in patients with LN was (9.11 ± 2.46) ng / mL, significantly higher than that (3.12 ± 0.97) ng / mL in non-LN patients and (1.12 ± 0.33) ng / mL in normal controls Significance (P <0.05). The mRNA levels of OX40 and OX40L in PBMCs of LN patients were (8 ± 4) × 106 copies / μL and (4 ± 3) × 103 copies / μL, respectively, significantly higher than those of non-LN patients [(4 ± 3) × 103 and (2 ± 2) × 103] and normal controls [(4 ± 2) × 103 and (2 ± 1) × 103], the difference was statistically significant (P <0.01). Conclusion The expressions of OX40 and OX40L mRNA in peripheral blood and anti-C1q antibody are correlated with LN activity, which may play a role in the occurrence and development of LN.
