目的:通过对HPVI6L1晚期蛋白的鉴定及电镜观察,为制备宫颈癌预防性疫苗奠定基础。方法:采用Westernblot方法对所表达蛋白进行特异性鉴定,并利用透射电镜观察HPVI6L1在酵母菌中自我组装成的病毒样颗粒(VLPs)。结果:所表达的外源蛋白能与鼠抗HPVI6L1单克隆抗体特异结合,证明该蛋白确为HPVI6L1基因编码,电镜下观察到了HPVI6L1的VLPs。结论:HPVI6L1基因经诱导可表达HPVI6L1晚期蛋白,该蛋白可自我组装成VLPs.
OBJECTIVE: Through the identification of HPVI6L1 late protein and electron microscopic observation, it lays the foundation for the preparation of preventive vaccine against cervical cancer. Methods: Western blotting was used to identify the expressed protein. Transmission electron microscopy was used to observe the self-assembled virus-like particles (VLPs) of HPVI6L1 in yeast. Results: The expressed foreign protein could specifically bind to mouse anti-HPVI6L1 monoclonal antibody, which proved that the protein was encoded by HPVI6L1 gene. The VLPs of HPVI6L1 were observed under electron microscope. Conclusion: HPVI6L1 gene can induce the expression of HPVI6L1 late protein, which can self-assemble into VLPs.
