目的调查浙江省啮齿动物巴尔通体携带状况,为巴尔通体人群感染的预防控制提供科学依据。方法用夹夜法在浙江省不同地区、不同季节捕获啮齿动物,无菌操作取啮齿动物的肝和脾,用聚合酶链反应(PCR)检测巴尔通体,然后对部分阳性产物测序,将所测核酸序列提交到GenBank,做相似性比较及序列分析。结果从黑线姬鼠、褐家鼠、小家鼠、东方田鼠、北社鼠和黄毛鼠中检测到巴尔通体特异DNA片段,阳性率分别为5.76%(17/295)、2.86%(1/35)、10.00%(1/10)、5.26%(1/19)、3.91%(7/179)和3.03%(1/33),浙江省首次从小家鼠中检测到巴尔通体DNA片段;岱山、海宁、建德、丽水、仙居、龙游地区的阳性率分别为2.63%(3/114)、10.84%(9/83)、2.88%(3/104)、11.11%(12/108)、0(0/46)和0.57%(1/175);4个季度阳性率分别为3.39%(4/118)、6.08%(9/148)、2.95%(7/237)、6.30%(8/127),平均阳性率为4.44%(28/630);遗传进化分析提示检测到的巴尔通体与Bartonella henselae遗传关系最接近。结论浙江省啮齿动物中广泛存在巴尔通体感染,存在对人群致病的风险,应采取措施加以控制。
Objective To investigate the carrying status of rodent Bartonella in Zhejiang Province and to provide a scientific basis for the prevention and control of Barr-infected population. Methods Rodents were harvested in different regions and seasons in Zhejiang Province by the method of clip-nocturna, and the rodent liver and spleen were aseptically manipulated. The Bartonella pallidum was detected by polymerase chain reaction (PCR), and then some positive products were sequenced. Nucleic acid sequences were submitted to GenBank for similarity comparison and sequence analysis. Results The specific DNA fragments of Barr-Bartonella were detected in Apodemus agrarius, Rattus norvegicus, Mus musculus, Microtus fortis, Beishekus and Rattus norvegicus, the positive rates were 5.76% (17/295), 2.86% (1 / 35), 10.00% (1/10), 5.26% (1/19), 3.91% (7/179) and 3.03% (1/33), respectively. The positive rates of Daishan, Haining, Jiande, Lishui, Xianju and Longyou were 2.63% (3/114), 10.84% (9/83), 2.88% (3/104) and 11.11% ), 0 (0/46) and 0.57% (1/175) respectively. The positive rates in 4 quarters were 3.39% (4/118), 6.08% (9/148), 2.95% (7/237) (8/127). The average positive rate was 4.44% (28/630). Genetic evolution analysis suggested that Bartonella henselae was the most closely related to Bartonella henselae. Conclusions There are widespread Bartonella in rodents in Zhejiang Province. There is a risk of pathogenicity to the population, and measures should be taken to control it.
