目的:对甘肃、陕西、宁夏、四川及青海5个主产地秦艽的遗传多样性进行分析。方法:用随机扩增多态性DNA标记(RAPD)技术对11个秦艽种群90个个体基因组DNA的遗传多样性进行检测。结果:RAPD 8条引物检测到65条扩增条带,其中有59条是多态的,多态位点百分比达到97.78%。通过聚类分析可将秦艽的11个种群聚为两类,四川和陕西两省4个种群聚为一类;甘肃、宁夏和青海省7个种群聚为一类。结论:甘肃省环县种群、子午岭种群和华亭种群应作为秦艽的重要种群加以优先重点保护。
Objective: To analyze the genetic diversity of Gentiana macrophylla in five main producing areas of Gansu, Shaanxi, Ningxia, Sichuan and Qinghai. Methods: The genetic diversity of genomic DNA in 90 individuals of 11 Gentiana macrophylla populations was detected by using random amplified polymorphic DNA (RAPD) technique. Results: Sixteen bands were detected by 8 RAPD primers, of which 59 were polymorphic and the percentage of polymorphic loci was 97.78%. Cluster analysis could cluster eleven populations of Gentiana macrophyllus into two groups, four clusters in Sichuan and Shaanxi provinces clustered into one category, and seven populations in Gansu, Ningxia and Qinghai provinces clustered into one category. Conclusion: The Huanxian population, Ziwuling population and Huating population of Gansu Province should be given priority protection as important populations of Gentiana macrophylla.
