目的探讨β-catenin(β-连环素)异常表达在原发性肝癌的发生、分化中的意义。方法西安航天总医院肝胆外科用免疫细胞化学检测体外培养的细胞株的β-catenin的表达,进一步研究β-catenin在原发性肝细胞癌的形成、分化中的作用。结果 WB细胞表面表达C-kit抗体,免疫细胞化学结果显示L2细胞株β-catenin几乎全部分布在细胞膜,细胞质、细胞核中几乎无表达,WB细胞株中β-catenin主要分布于细胞膜、细胞质,细胞核中有较少表达,HepG2、SMCC细胞株中β-catenin几乎全部分布于细胞质、细胞核中并有较高表达,SMCC表达多于HepG2细胞株。western blot定量显示β-catenin在L2、WB、HepG2、SMCC细胞株的细胞膜表达逐渐减少,细胞质、细胞核中的表达逐渐增多。结论β-catenin由细胞膜逐渐向细胞质、细胞核的异位表达在原发性肝癌的发生、分化过程中可能具有重要作用。
Objective To investigate the significance of abnormal expression of β-catenin in the occurrence and differentiation of primary liver cancer. Methods Immunocytochemistry of hepatobiliary surgery in Xi’an Aerospace General Hospital was used to detect the expression of β-catenin in cell lines cultured in vitro to further investigate the role of β-catenin in the formation and differentiation of primary hepatocellular carcinoma. Results The C-kit antibody was expressed on the surface of WB cells. The results of immunocytochemistry showed that almost all the β-catenin in L2 cell line was distributed in the cell membrane with little expression in the cytoplasm and nucleus. The β-catenin in WB cell line mainly distributed in the cell membrane, cytoplasm and nucleus The expression of β-catenin in HepG2 and SMCC cell lines was almost completely distributed in the cytoplasm and nucleus, and was higher in HepG2 and SMCC than in HepG2. Western blot quantitative analysis showed that the expression of β-catenin in the cell membrane of L2, WB, HepG2 and SMCC cells gradually decreased, and the expression of β-catenin gradually increased in cytoplasm and nucleus. Conclusion The ectopic expression of β-catenin from cytoplasm to cytoplasm and nucleus may play an important role in the occurrence and differentiation of primary hepatocellular carcinoma.
