目的探讨益气开秘方对结肠慢传输型便秘小鼠肠道动力的影响。方法将30只小鼠分为正常组、模型组和中药组,后两组采用吗啡皮下注射制备小鼠肠动力障碍模型;正常组常规饲养,模型组灌胃给予0.9%NaCl溶液,中药组灌胃给予益气开秘方;4周后检测炭末推进百分率,以及结肠组织血管活性肠肽(VIP)、结肠Cajal细胞标志物C-Kit蛋白表达。结果模型组炭末推进率较正常组明显降低(P<0.05),中药组与模型组比较,炭末推进百分率明显提高,差异有统计学意义(P<0.05)。模型组VIP阳性神经纤维较正常组降低,中药组较模型组明显提高,差异有统计学意义(P<0.05)。模型组C-Kit蛋白表达较正常组减少,中药组较模型组有明显增加,差异有统计学意义(P<0.05)。结论益气开秘方对吗啡诱导肠动力障碍小鼠具有一定肠动力恢复作用,其可能通过促进结肠VIP表达、修复Cajal间质细胞,发挥调控肠道动力作用。
Objective To investigate the effect of Yiqi Kaimi Decoction on intestinal motility in slow transit constipation mice. Methods 30 mice were divided into normal group, model group and traditional Chinese medicine group. The latter two groups were injected morphine subcutaneously to establish model of intestinal motility disorder. Normal group were fed with 0.9% NaCl solution by intragastric administration. After 4 weeks, the percentage of charcoal propulsion and the expression of vasoactive intestinal peptide (VIP) in colon tissue and C-Kit protein in colon Cajal cell were detected. Results Compared with normal group, the advancing rate of carbon powder in model group was significantly lower than that in normal group (P <0.05). Compared with model group, the propulsion rate of charcoal significantly increased (P <0.05). The VIP-positive nerve fibers in the model group were lower than those in the normal group, and were significantly increased in the TCM group compared with the model group (P <0.05). The protein expression of C-Kit in the model group decreased compared with that in the normal group, but the difference was statistically significant (P <0.05). Conclusions Yiqi Kaimi Recipe has a certain intestinal motility recovery effect on morphine-induced gut dysmotility mice, which may play a role in regulating intestinal motility by promoting VIP expression of colon and repairing Cajal interstitial cells.
