目的:探讨补肾通窍方对糖尿病大鼠下丘脑室旁核神经元Bax与Bcl-2表达的影响。方法:腹腔内注射链脲佐菌素(streptozotocin,STZ)55mg/kg制作糖尿病大鼠模型,采用免疫组化SABC法检测各组大鼠下丘脑室旁核神经元Bax与Bcl-2的表达。结果:①糖尿病模型组Bax阳性细胞数较正常对照组增多(P<0.01),Bcl-2阳性细胞数减少(P<0.01)。②补肾通窍方治疗组和胰岛素治疗组Bax阳性细胞数比糖尿病模型组减少(P<0.01),而Bcl-2阳性细胞数则增多(P<0.01)。③补肾通窍方治疗组和胰岛素治疗组的Bax和Bcl-2阳性细胞数无显著差异(P>0.05)。结论:补肾通窍方能抑制糖尿病大鼠下丘脑室旁核神经元Bax的表达,促进Bcl-2的表达,与胰岛素相比哪者效果更好尚待研究。
Objective: To investigate the effect of Bushen Tongqiao decoction on Bax and Bcl-2 expression in hypothalamic paraventricular nucleus neurons in diabetic rats. Methods: Diabetic rat models were induced by intraperitoneal injection of streptozotocin (STZ) 55 mg / kg. The expression of Bax and Bcl-2 in neurons of hypothalamic paraventricular nucleus was detected by immunohistochemical SABC method. Results: ①The number of Bax positive cells in diabetic model group was higher than that in normal control group (P <0.01), and the number of Bcl-2 positive cells decreased (P <0.01). ② The number of Bax positive cells in Bushen Tongqiao decoction group and insulin treatment group was lower than that in diabetic model group (P <0.01), while the number of Bcl-2 positive cells increased (P <0.01). ③ There was no significant difference in Bax and Bcl-2 positive cells between Bushen Tongqiao decoction group and insulin treatment group (P> 0.05). Conclusion: Bushen Tongqiao decoction can inhibit the expression of Bax in hypothalamic paraventricular nucleus neurons and promote the expression of Bcl-2 in diabetic rats, which is better than insulin.
