目的:探讨黄连素诱导人肝癌HepG2细胞凋亡的作用及其机制。方法:不同浓度的黄连素处理人肝癌HepG2细胞,采用MTT法检测黄连素对HepG2细胞的抑制率,用流式细胞仪检测黄连素对HepG2细胞凋亡的影响,Western blot法检测细胞p53和BCL-2蛋白的表达。结果:黄连素可显著的降低HepG2细胞的存活率,流式细胞仪表明在黄连素作用48h后,HepG2细胞的凋亡率显著增加,同时给药组细胞中的p53蛋白表达显著升高,BCL-2蛋白表达显著降低。结论:黄连素可通过上调p53蛋白表达、下调BCL-2蛋白表达来诱导HepG2细胞凋亡。
Objective: To investigate the effect of berberine on inducing apoptosis of HepG2 cells and its mechanism. Methods: HepG2 cells were treated with different concentrations of berberine. The inhibitory rates of berberine on HepG2 cells were measured by MTT assay. The apoptosis of HepG2 cells was detected by flow cytometry. The expressions of p53 and BCL -2 protein expression. Results: Berberine can significantly reduce the survival rate of HepG2 cells. Flow cytometry showed that the apoptosis rate of HepG2 cells increased significantly after 48 hours of berberine treatment, and the expression of p53 protein in BCL -2 protein expression was significantly reduced. Conclusion: Berberine can induce HepG2 cell apoptosis by up-regulating the expression of p53 protein and down-regulating the expression of BCL-2 protein.
