RAPD是一种新的分子生物学技术。本实验用28个随机引物对平菇、香菇及其融合子进行PCR扩增,在融合子谱带中,有64条与平菇相同(与平菇的相似指数SI=70.6%),14条与香菇相同(SI=33.6%),另外有26条谱带为双亲与融合子所共有。本实验结果说明融合子是双亲融合的产物,但在融合过程中,从双亲获得的遗传信息不等。本实验首次在食用真菌中获得了原生质体融合的分子生物学证据。我们认为,在食用菌研究中,RAPD将是一种有重要价值的分析方法。
RAPD is a new molecular biology technique. In this experiment, 28 random primers were used for PCR amplification of Pleurotus ostreatus, Mushrooms and their fusions. Among them, 64 were identical to Pleurotus ostreatus (similarity index SI = 70.6%) with Pleurotus ostreatus, 14 were the same as shiitake mushrooms (SI = 33.6%), and another 26 were shared by both parents and fusants. The experimental results show that the fusion is the product of the parental fusion, but the genetic information obtained from both parents during the fusion process varies. In this experiment, the molecular biological evidence of protoplast fusion was obtained for the first time in edible fungi. In our opinion, RAPD will be a valuable analytical method in edible fungi research.
