目的:采用HPLC法监测癫痫患者体内卡马西平和苯妥英钠的血药浓度。方法:收集同时服用卡马西平与苯妥英钠的癫痫患者的血样作为研究对象,经二氯甲烷提取,流动相为甲醇:水=50∶50;色谱柱:InertSustain-C18(250×4.6mm,5μm);流速为1.0ml·min-1;柱温:25℃;紫外检测波长:240nm。结果:卡马西平的-苯妥英钠的线性范围分别为和2.35~28.31μg·mL-1和1.87~22.47μg·mL-1定量限分别为卡马西平2.35μg·mL-1,苯妥英钠1.87μg·mL-1。两者的低、中、高回收率均在85.2%~93.6%之间,其日间日内标准差均在2%以内。结论:本方法稳定、快速、灵敏度高、专属性强,可用于常规治疗药物检测及卡马西平及苯妥英钠体内代谢性研究。
OBJECTIVE: To monitor plasma concentrations of carbamazepine and phenytoin sodium in patients with epilepsy by HPLC. Methods: The blood samples from patients with epilepsy who took carbamazepine and phenytoin sodium simultaneously were collected and extracted with dichloromethane. The mobile phase was methanol: water = 50:50. Column: Inertustain-C18 (250 × 4.6mm, 5μm ); Flow rate 1.0ml · min-1; column temperature: 25 ℃; UV detection wavelength: 240nm. RESULTS: The linear range of carbamazepine-phenytoin sodium was 2.35 ~ 28.31μg · mL-1 and 1.87 ~ 22.47μg · mL-1, respectively. The limits of quantification were 2.35μg · mL-1 for carbamazepine, 1.87μg for phenytoin · ML-1. Both low, medium and high recovery rates were between 85.2% to 93.6%, the daytime standard deviations are within 2%. Conclusion: The method is stable, rapid, sensitive and specific. It can be used in routine therapy and in vivo metabolism of carbamazepine and phenytoin.
