【目的】比较接触性超敏反应(CHS)小鼠不同部位CD4+/CD8+T细胞以及辅助性T细胞(Th1/Th2)、细胞毒性T细胞(Tc1/Tc2)的平衡状态。【方法】以二硝基氟苯(DNFB)致敏和攻击BALB/c小鼠,建立CHS模型。采用三色流式细胞术分析CHS小鼠脾脏(SP)、耳后淋巴结(RLN)、小肠派氏结(PPs)和肠系膜淋巴结(MLN)中CD4+T细胞和CD8+T细胞的变化及其胞内细胞因子干扰素-γ(IFN-γ)与白细胞介素-4(IL-4)的表达水平,观察CHS小鼠不同部位Th1/Th2、Tc1/Tc2比例的变化。【结果】与正常组比较,CHS组RLN与SP中CD4/CD8比值显著下降(均P<0.01),RLN中以Tc1与Th1占优势,SP中以Tc1与Th2占优势,PPs中Tc1细胞比例、CD4/CD8比值显著上升(均P<0.05),MLN中各指标无显著性差异。【结论】DNFB诱发的CHS小鼠机体总体免疫平衡向Th1类偏移,Tc1(CD8+IFN-γ+)细胞是主要的效应细胞。
【Objective】 To compare the equilibrium status of CD4 + / CD8 + T cells and T helper (Th1 / Th2) and cytotoxic T cells (Tc1 / Tc2) in different parts of contact hypersensitivity (CHS) mice. 【Method】 BALB / c mice were sensitized and challenged with dinitrofluorobenzene (DNFB) to establish CHS model. The changes of CD4 + T cells and CD8 + T cells in spleen (SP), posterior ear lymph nodes (RLN), small intestine sentinel lymph nodes (PPs) and mesenteric lymph nodes (MLN) (IFN-γ) and interleukin-4 (IL-4) levels in CHS mice were observed. The changes of Th1 / Th2 and Tc1 / Tc2 ratios in different parts of CHS mice were observed. 【Results】 Compared with the normal group, the ratio of CD4 / CD8 in RLN and SP in CHS group was significantly decreased (all P <0.01), Tc1 and Th1 in RLN were predominant, while Tc1 and Th2 in SP were predominant. The proportion of Tc1 in PPs , CD4 / CD8 ratio increased significantly (all P <0.05), MLN no significant difference in each index. 【Conclusion】 The overall immune balance of CHS mice induced by DNFB is shifted to Th1 class. Tc1 (CD8 + IFN-γ +) cells are the main effector cells.
