目的建立双氧水(H_2O_2)诱导人肠黏膜上皮细胞(Caco-2)细胞的溃疡性结肠炎细胞模型,通过观察猴头菌多糖对该细胞模型影响,探讨其与氧化应激相关的抗溃疡性结肠作用机制。方法通过CCK-8法检测对细胞模型增殖的影响;通过Elisa法检测对细胞内超氧化物歧化酶(SOD)、丙二醛(MDA)、乳酸脱氢酶(LDH)的含量变化;通过流式细胞术检测细胞凋亡率的影响。结果与细胞模型组相比较,猴头菌多糖能明显增加H_2O_2诱导的Caco-2细胞增殖、明显降低细胞凋亡率,其细胞中MDA含量明显降低(P<0.05),SOD及LDH含量明显提高(P<0.05)。结论猴头菌多糖具有抗H_2O_2引起Caco-2细胞凋亡的作用,提示其抗溃疡性结肠炎活性可能与减少氧化应激相关。
Objective To establish a model of ulcerative colitis cells induced by hydrogen peroxide (H_2O_2) in human intestinal epithelial cells (Caco-2). To observe the effects of Hericium erinaceus polysaccharides on the cell model, and to investigate its anti-ulcerative colitis Mechanism. Methods The effects of CCK-8 on cell proliferation were detected. The content of superoxide dismutase (SOD), malondialdehyde (MDA) and lactate dehydrogenase (LDH) in cells were detected by Elisa method. Cytometry was used to detect the effect of apoptosis rate. Results Compared with the model group, the polysaccharide of Hericium erinaceus could significantly increase the proliferation of Caco-2 cells induced by H 2 O 2 and significantly reduce the apoptosis rate of cells. The content of MDA in the cells was significantly decreased (P <0.05) and the content of SOD and LDH was significantly increased (P <0.05). Conclusion Hericium erinaceus polysaccharides have anti-H_2O_2-induced apoptosis in Caco-2 cells, suggesting that the anti-ulcerative colitis activity may be related to the reduction of oxidative stress.
