目的通过检测胃腺癌组织中NF-κBp65、VEGF、Survivin的表达情况,探讨它们在胃腺癌发生发展过程中的作用以及相互作用关系。方法应用免疫组织化学SP法,检测73例胃癌组织及20例正常胃黏膜组织中NF-κBp65、VEGF、Survivin的表达情况,并分析其与胃癌临床病理特征之间的关系。结果NF-κBp65、VEGF、Survivin在胃腺癌中的表达率分别为58.9%、72.6%、46.6%,而在正常胃黏膜组织则表达不明显或不表达,两者差异有统计学意义(P<0.05)。NF-κBp65、VEGF、Survivin的表达与浸润深度、淋巴结转移、临床分期均密切相关(P<0.05),NF-κBp65的表达亦与胃腺癌分化程度有关(P<0.05)。NF-κBp65蛋白的表达与VEGF及Survivin蛋白的表达均呈显著正相关,VEGF与Survivin的表达亦呈显著正相关。结论NF-κBp65通过上调VEGF及Survivin的表达促进胃腺癌的发生发展;VEGF与Survivin在胃腺癌的发生发展中存在着协同作用。
Objective To detect the expression of NF-κBp65, VEGF and Survivin in gastric adenocarcinoma and to explore their roles in the development and progression of gastric adenocarcinoma. Methods The expressions of NF-κBp65, VEGF and Survivin in 73 cases of gastric cancer and 20 cases of normal gastric mucosa were detected by immunohistochemical SP method. The relationship between NF-κBp65, VEGF, Survivin and the clinicopathological features of gastric cancer was analyzed. Results The positive rates of NF-κBp65, VEGF and Survivin in gastric adenocarcinoma were 58.9%, 72.6% and 46.6% respectively, but not significantly or not in normal gastric mucosa Statistical significance (P <0.05). The expression of NF-κBp65, VEGF and Survivin was closely related to the depth of invasion, lymph node metastasis and clinical stage (P <0.05). The expression of NF-κBp65 also correlated with the differentiation of gastric adenocarcinoma (P <0.05). The expression of NF-κBp65 protein was positively correlated with the expression of VEGF and Survivin protein. There was also a significant positive correlation between the expression of VEGF and Survivin. Conclusion NF-κBp65 promotes the development of gastric adenocarcinoma by up-regulating the expression of VEGF and Survivin. There is synergistic effect between VEGF and Survivin in the development of gastric adenocarcinoma.
