目的建立土茯苓中总多糖的含量测定方法,对32个不同采集地切面红棕色及类白色土茯苓中的总多糖含量进行比较分析。方法采用紫外-可见分光光度法,以葡萄糖为对照品,用苯酚-浓硫酸法显色,于490 nm处测定样品溶液中的吸光度,计算出样品中总多糖的含量。结果通过对土茯苓多糖含量测定多个环节的考察,建立了土茯苓中总多糖的含量测定方法。该方法对多糖的提取率高,得标准曲线:A=9.7853X-0.0473,r=0.9990(n=7),葡萄糖在9.988~119.856μg之间有良好的线性关系。平均加样回收率为102.2%,RSD为2.4%(n=6)。结论其它省区切面类白色土茯苓总多糖的含量明显高于贵州各产地切面红棕色土茯苓的含量,为土茯苓的质量控制提供了一定的依据。
Objective To establish a method for the determination of total polysaccharide in Smilax glabra by comparing the content of total polysaccharides in the reddish brown and white tilapia (Triticum aestivum L.) collected from 32 different places. Methods Ultraviolet-visible spectrophotometry was used to determine the total polysaccharides in the sample with glucose as reference substance and phenol-concentrated sulfuric acid method. The absorbance in the sample solution was measured at 490 nm. Results Through the examination of many aspects of the determination of Smilax Polysaccharide, the method of determination of total polysaccharide in Smilax Glabra was established. The method of polysaccharide extraction rate, the standard curve: A = 9.7853X-0.0473, r = 0.9990 (n = 7), glucose in the 9.988 ~ 119.856μg between a good linear relationship. The average recovery was 102.2% with a RSD of 2.4% (n = 6). Conclusion The contents of total polysaccharides of the white tuckahoe in the other provinces and autonomous regions are obviously higher than those of Poria cocos in the reddish brown soil of all producing areas in Guizhou Province, which provides a basis for the quality control of tuckahoe.
