研究马尾松针叶聚戊烯醇体外和体内的抗氧化活性,体外测定其对羟自由基(·OH)、超氧阴离子自由基(O2-·)和二苯基苦基苯肼自由基(DPPH·)的清除能力,以及对肝匀浆和肝线粒体自发性及Fe2+诱导性脂质过氧化的抑制作用,体内测定大鼠血清、肝匀浆和心匀浆超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性和丙二醛(MDA)含量变化。结果表明:马尾松针叶聚戊烯醇能够有效抑制大鼠肝匀浆和肝线粒体脂质过氧化及肝线粒体肿胀程度,清除·OH、O-2.和DPPH·半数抑制浓度(IC50)分别为34.87、27.13、58.23 mg/L,增强大鼠血清、肝脏和心脏中SOD、GSH-Px活性和降低MDA含量(p<0.05)。马尾松针叶聚戊烯醇在体外和体内具有明显的抗氧化活性,为马尾松针叶聚戊烯醇在药物中的应用提供依据。
In vitro and in vivo antioxidant activity of Pinus massoniana coniferous seed puerarin was investigated in vitro. The effects of hydroxyl radical (· OH), superoxide anion radical (O2- ·) and diphenyl picrylhydrazyl radical (DPPH ·), As well as the inhibitory effect on liver homogenate and hepatic mitochondria spontaneous and Fe2 + -induced lipid peroxidation. The levels of superoxide dismutase (SOD) in serum, liver homogenate and cardiac homogenate were determined in vivo, Glutathione peroxidase (GSH-Px) activity and malondialdehyde (MDA) content. The results showed that P. massoniana needles could effectively inhibit lipid peroxidation and mitochondria swelling in rat liver homogenates and hepatic mitochondria, and the concentrations of · OH, O-2 · and DPPH · half-inhibitory concentrations (IC50) were 34.87,27.13,58.23 mg / L, respectively. The activity of SOD and GSH-Px in serum, liver and heart were increased and MDA content was decreased (p <0.05). Masson pine needles polyprenols have obvious antioxidant activity in vitro and in vivo, providing the basis for the application of Pinus massoniana coniferol polyprenols in medicine.
