目的:探讨蛋白酪氨酸磷酸酶SHP-1蛋白在γ射线体外诱发的白血病小鼠胸腺细胞中的变化特点。方法:建立γ射线体外诱发BALB/c小鼠白血病模型。实验分为癌变组、未癌变组及对照组,从各组小鼠获取胸腺细胞,应用Western印迹、免疫沉淀法及生物化学方法检测各组胸腺细胞SHP-1蛋白质的表达、酪氨酸磷酸化水平及酶活性。结果:癌变组SHP-1蛋白的表达明显高于对照组及未癌变组(P<0.01),但各组SHP-1的酪氨酸磷酸化水平及酶催化活性差异不显著(P>0.05),且其酪氨酸磷酸化水平与酶活性间存在明显相关性。结论:在γ射线诱发的白血病小鼠的胸腺细胞中,SHP-1蛋白表达明显升高,但可能存在酪氨酸磷酸化障碍,导致其活性降低,即表现出SHP-1蛋白表达与其活性分离现象,提示辐射致癌中SHP-1蛋白可能存在功能障碍。
Objective: To investigate the changes of protein tyrosine phosphatase SHP-1 protein in thymocytes of leukemia mice induced by γ-rays in vitro. Methods: Establishing γ ray to induce BALB / c mouse leukemia model in vitro. The experiment was divided into canceration group, non-cancerous group and control group. Thymocytes were obtained from mice in each group. Western blotting, immunoprecipitation and biochemical methods were used to detect the expression of SHP-1 protein, tyrosine phosphorylation Level and enzyme activity. Results: The SHP-1 protein expression in canceration group was significantly higher than that in control group and non-cancerous group (P <0.01), but there was no significant difference in SHP-1 tyrosine phosphorylation level and enzyme catalytic activity in each group (P> 0.05) , And there is a clear correlation between tyrosine phosphorylation level and enzyme activity. CONCLUSIONS: SHP-1 protein expression is significantly increased in γ-ray-induced leukemic thymocytes but may be associated with an impaired tyrosine phosphorylation resulting in a decrease in SHP-1 protein expression and its activity Phenomenon, suggesting that radiation-induced SHP-1 protein may have dysfunction.
