归脾汤对苯中毒小鼠血清中CD_3、CD_4和CD_8的影响

目的观察归脾汤对苯中毒小鼠T细胞亚群水平的影响。方法 72只昆明小鼠随机分为正常对照组、模型组、西药组以及归脾汤大、中、小剂量组,每组12只。将小鼠置于人工模拟高苯浓度环境每天8 h,连续14天,复制苯中毒模型。造模后,归脾汤大、中、小剂量组分别给予归脾丸溶液8、4、2 mg/(kg·d)灌胃;西药组给予利血生1.5 mg/(kg·d)和鲨肝醇5 mg/(kg·d)混悬液灌胃;正常对照组和模型组给予生理盐水灌胃,均每天1次,连续3周。用全自动血细胞分析仪、流式细胞仪分别检测小鼠外周血象、外周血T淋巴细胞亚群CD3、CD4和CD8计数的变化,并计算胸腺及脾脏指数。结果归脾汤大、小剂量组的胸腺重量、胸腺指数均增加,差别均有统计学意义(P<0.05)。归脾汤小剂量组的脾脏及脾脏指数均增加差别有统计学意义(P<0.05)。归脾汤各组及西药组CD4+、CD8+和CD4/CD8的表达均上升,且差别均有统计学意义(P<0.05)。结论归脾汤可引起苯中毒小鼠外周血T淋巴细胞亚群CD4+、CD8+增加,从而使免疫功能增强。 Objective To observe the effect of Guipi Decoction on T lymphocyte subsets in benzene poisoning mice. Methods Totally 72 Kunming mice were randomly divided into normal control group, model group, western medicine group and Guipi Decoction group, with 12 rats in each group. The mice were placed in artificial simulated high benzene concentration environment 8 h per day, continuous 14 days, replication of benzene poisoning model. After modeling, the Guipi Decoction large, medium and small dose groups were given Guipi Pill solution 8,4,2 mg / (kg · d) gavage; western medicine group was given reserpine 1.5 mg / (kg · d) and Shark liver alcohol 5 mg / (kg · d) suspension of intragastric administration; normal control group and model group were given physiological saline gavage once a day for 3 weeks. The changes of counts of CD3, CD4 and CD8 in peripheral blood T lymphocyte subsets and peripheral blood were detected by automatic hematology analyzer and flow cytometry, respectively. Thymus and spleen indexes were calculated. Results Guipi Tang large and small dose group of thymus weight, thymus index were increased, the difference was statistically significant (P <0.05). Guipi Decoction low dose group of spleen and spleen index were significantly different (P <0.05). The expressions of CD4 +, CD8 + and CD4 / CD8 in Guipi Decoction group and western medicine group all increased, and the differences were statistically significant (P <0.05). Conclusion Guipi Decoction can cause peripheral blood T lymphocyte subsets CD4 + and CD8 + in benzene poisoning mice to increase, so that immune function is enhanced.
