目的:研究补肾活血方对免疫性卵巢早衰小鼠PI3、Akt、Bcl2蛋白表达的影响,为临床治疗卵巢早衰提供思路。方法:以小鼠ZP3为抗原,皮下多点注射免疫BALB/c雌性小鼠建立免疫性POF模型。以补肾活血方低、中、高剂量进行治疗,同步设阳性对照组(补佳乐)和空白组(生理盐水)。给予相应药物。治疗21 d后采用HE染色法检测卵巢组织的病理变化,免疫组化方法检测卵巢颗粒细胞PI3K、Akt、Bcl2蛋白表达,并分析阳性表达的平均光密度。结果:模型组PI3K、Akt和Bcl2蛋白表达量明显减低,与空白组比较,差异具有统计学意义(P<0.01);各药物组干预后PI3K、Akt和Bcl-2蛋白表达量明显上调,与模型组比较,差异具有统计学意义(P<0.01);其中阳性药物组和高剂量组表达上调最为明显,且两组的效果等同。结论:补肾活血方改善卵巢功能的机制可能与上调颗粒细胞PI3、Akt、Bcl2的蛋白表达有关。
Objective: To study the effects of Bushen Huoxue Recipe on PI3, Akt and Bcl2 protein expression in premature ovarian failure mice and to provide ideas for clinical treatment of premature ovarian failure. Methods: BALB / c female BALB / c mice were injected subcutaneously with mouse ZP3 as an antigen to establish an immune POF model. To Bushen Huoxue Fang low, medium and high doses for treatment, set simultaneously positive control group (nadir) and the blank group (saline). Give the appropriate medicine. After 21 days of treatment, the pathological changes of ovarian tissue were detected by HE staining. The protein expressions of PI3K, Akt and Bcl2 in ovarian granulosa cells were detected by immunohistochemistry and the average optical density of positive expression was analyzed. Results: The expression of PI3K, Akt and Bcl2 in the model group was significantly lower than that in the blank group (P <0.01), and the expressions of PI3K, Akt and Bcl-2 were significantly up-regulated The difference between the model group and the high dose group was statistically significant (P <0.01). The positive group and the high dose group were most obviously up-regulated, and the effect of the two groups was the same. Conclusion: The mechanism of Bushen Huoxue Decoction in improving ovarian function may be related to up-regulating the protein expression of PI3, Akt and Bcl2 in granulosa cells.
