目的:研究桂竹糖芥苷G(erysimine G)的利尿作用及其作用机制。方法:应用尿道插管观察给药前后尿量变化;并应用膜片箝技术记录大鼠肾脏髓袢升支粗段70-pS钾通道的活性改变。结果:给予桂竹糖芥苷G后,实验动物尿量显著增加。单通道实验记录表明:应用桂竹糖芥苷G1μmol·L~(-1)抑制TAL70-pS钾通道的活性,NP_o由正常对照的1.67±0.10降低至0.05±0.02(P<0.01,n=4)。结论:桂竹糖芥苷G有明显的利尿作用,其利尿机制可能与抑制TAL单细胞70-pS钾通道的活性有关。
Objective: To study the diuretic effect and its mechanism of action of erysimillin G in bamboo. Methods: Urinary changes were observed before and after administration by urethral catheterization. The activity of 70-pS potassium channels in the ascending and descending segments of the rat spinal cord was recorded using patch clamp technique. Results: The urine output of experimental animals was significantly increased after administration of caneraditoside G. A single-channel experiment indicated that the activity of TAL70-pS potassium channel was inhibited by using G1 bamboo polysaccharide glucoside G1 μmol·L-1, and the NP_o was decreased from 1.67±0.10 to 0.05±0.02 in normal controls (P<0.01, n=4). . Conclusion: Glycosaminoglycan G has obvious diuretic effect, and its diuretic mechanism may be related to inhibiting the activity of TAL single-cell 70-pS potassium channel.
