目的:建立液相色谱法测定燕窝中唾液酸含量的方法。方法:300SCX阳离子交换色谱柱,4.6×250mm,粒径5μm,流动相:乙腈-0.1%磷酸水溶液(90:10),柱温:30℃,流速:1.0ml/min,进样量:10μl,检测波长:205nm。结果:系统研究了唾液酸在不同色谱柱中的保留行为,确定SCX型阳离子交换柱对唾液酸有最好的保留;线性范围:4~100μg/ml,线性方程:y=5.08517x+0.0023224,r=0.99996;燕窝平均回收率为96.8%,RSD=1.16%;冰糖燕窝平均回收率为96.4%,RSD=1.00%。结论:本法可用于燕窝及燕窝制品的质量控制。
Objective: To establish a method for the determination of sialic acid in bird’s nest by liquid chromatography. Method: 300 × 300mm cation exchange column, 4.6 × 250mm, particle size 5μm, mobile phase: acetonitrile-0.1% phosphoric acid solution 90:10, Detection wavelength: 205nm. Results: The retention behavior of sialic acid in different columns was systematically studied. The best retention of sialic acid was confirmed by SCX cation exchange column. The linear range was 4-100μg / ml, the linear equation was y = 5.08517x + 0.0023224, r = 0.99996. The average recovery of bird’s nest was 96.8% and RSD was 1.16%. The average recovery of rock sugar bird’s nest was 96.4% and RSD was 1.00%. Conclusion: This method can be used for quality control of bird’s nest and bird’s nest products.
