目的:通过预实验观察化痰祛湿活血方对非酒精性脂肪性肝炎(NASH)大鼠血清及肝组织中相关氧化应激因子的影响,初步探讨其治疗NASH的作用机制。方法:SPF级SD雄性大鼠随机分为空白组、模型组、对照组、高剂量组、中剂量组、低剂量组,空白组予普通饲料,其余组给予高脂饲料联合四环素腹腔注射造模。造模第9周处死全部实验动物,HE染色观察肝组织病理结构,ELISA法测定血清及肝组织中MCP-1、PTGS2、NOS2、MDA含量。结果:各用药组血清及肝组织中MCP-1、PTGS、NOS2、MDA含量均低于模型组(P<0.01或P<0.05),高剂量组降低最显著,优于对照组(P<0.05)。结论:化痰祛湿活血方可减轻NASH大鼠炎症损伤,其机制可能与其能够下调氧化应激相关因子MCP-1、PTGS2、NOS2、MDA表达有关。
OBJECTIVE: To investigate the effect of Huatan Huashi Huoxue Recipe on serum and liver tissue related oxidative stress factors in nonalcoholic steatohepatitis (NASH) rats and to explore its mechanism of action in NASH. Methods: SPF SD male rats were randomly divided into normal group, model group, control group, high dose group, middle dose group, low dose group and blank group. The other groups were given high fat diet combined with tetracycline intraperitoneal injection . At the 9th week, all the experimental animals were sacrificed, the pathological structure of liver was observed by HE staining, and the levels of MCP-1, PTGS2, NOS2 and MDA in serum and liver were measured by ELISA. Results: The contents of MCP-1, PTGS, NOS2 and MDA in serum and liver of each treatment group were lower than those in model group (P <0.01 or P <0.05), and those in high dose group were the most significant ). Conclusion: Huatan Qushi Huoxue Recipe can reduce the inflammatory injury in NASH rats, and its mechanism may be related to its ability to down-regulate the expression of oxidative stress factors MCP-1, PTGS2, NOS2 and MDA.
