目的用温阳解郁三种不同组方对模拟早年不良经历的母婴分离小鼠进行干预,比较其治疗效果并探讨其作用机理。方法选用从出生起连续10天、每天3h母婴分离的12~20周成年C57BL/6J小鼠遭受社会击败应激塑造抑郁症模型。分别以逍遥散、二仙汤和温阳解郁复方进行6周的干预,通过旷场、高架十字迷宫及强迫游泳实验观察行为模式变化并运用原位杂交检测不同脑区BDNF的表达。结果模型小鼠在旷场实验中央区域的活动减少,高架十字迷宫实验中开放臂和中央交汇区进入次数及停留时间均明显减少,强迫游泳不动时间明显延长,提示该模型已有明显的抑郁或焦虑表现。二仙汤和温阳解郁复方均能显著提高中央交汇区进入次数(P<0.05),温阳解郁复方能显著缩短强迫游泳不动时间(P<0.05)。除前额皮质(PFC)外,模型小鼠各脑区的BDNF表达均明显下降,各治疗组均能明显提高各脑区BDNF mRNA的表达量。结论温阳解郁复方对抑郁症有明显治疗作用,可能是温阳和解郁互相配合的结果。
OBJECTIVE To intervene three different groups of mice, WenYangXieYu, to separate the maternal and infant mice, which had bad experiences in the early years, and to compare their therapeutic effects and explore the mechanism of action. METHODS: Adult C57BL / 6J mice aged 12 to 20 weeks separated from their mothers and their infants for 10 days from birth were subjected to social defeat stress to model depression. The interventions of Xiaoyaosan, Erxian Decoction and Wenyang Jieyu Compound were performed for 6 weeks respectively. The changes of behavioral patterns were observed by open field, elevated plus maze and forced swimming test, and the expression of BDNF in different brain regions was detected by in situ hybridization. Results The model mice showed less activity in the central area of open-field test. In the elevated maze test, the entry times and stay times of open arms and central intersections were significantly decreased, forced swimming time was significantly prolonged, suggesting that the model had obvious depression Or anxiety performance. Erxian Decoction and Wenyang Jieyu Compound can significantly increase the number of entry into the central junction (P <0.05). Wenyang Jieyu Compound can significantly reduce forced swimming time (P <0.05). In addition to prefrontal cortex (PFC), BDNF expression was significantly decreased in all brain regions of model mice, and the expression of BDNF mRNA in each brain region was significantly increased in all treatment groups. Conclusions Wenyang Jieyu Compound has a significant therapeutic effect on depression, which may be the result of the combination of Wenyang and Xieyu.
