目的研究Wortmannin对急性肺损伤模型小鼠肺组织白介素-6(IL-6)和白介素-10(IL-10)表达的影响。方法30只昆明小鼠随机分为正常对照组、急性肺损伤组和Wortmannin处理组。采用腹腔注射LPS(10 mg/kg)建立小鼠急性肺损伤模型,对照组腹腔注射同体积的生理盐水,Wortmannin处理组则于造模前2 h腹腔注射Wortmannin(1.4 mg/kg)。LPS注射后6 h处死大鼠,计算肺组织湿/干重(w/D)比值,Western blot方法检测三组小鼠肺组织内IL-6和IL-10的表达变化,RT-PCR方法检测三组小鼠肺组织内IL-6 mRNA和IL-10 mRNA的表达变化。结果急性肺损伤组小鼠肺组织IL-6蛋白表达显著上升,平均光密度值为(0.38±0.03),显著高于正常对照组的表达(0.09±0.01),P<0.05;相比于急性肺损伤组小鼠,Wortmannin处理组小鼠肺组织IL-6蛋白表达显著降低,光密度值为(0.21±0.02),P<0.05;急性肺损伤组小鼠肺组织IL-10蛋白表达显著降低,平均光密度值为(0.11±0.01),显著低于正常对照组的表达(0.21±0.01),P<0.05;相比于急性肺损伤组小鼠,Wortmannin处理组小鼠肺组织IL-10蛋白表达显著升高,光密度值为(0.18±0.02),P<0.05。RT-PCR方法检测三组小鼠IL-6mRNA时发现,Wortmannin处理组小鼠肺组织IL-6 mRNA的平均光密度值为(0.37±0.03),显著低于急性肺损伤组小鼠肺组织IL-6 mRNA的的平均光密度值为(0.26±0.02),P<0.05;RT-PCR方法检测三组小鼠IL-10mRNA时发现,Wortmannin处理组小鼠肺组织IL-10 mRNA的平均光密度值为(0.27±0.02),显著高于急性肺损伤组小鼠肺组织IL-10 mRNA(0.18±0.02)的表达,P<0.05。结论 Wortmannin能抑制急性肺损伤小鼠肺组织IL-6表达,上调IL-10的表达。
Objective To investigate the effects of Wortmannin on the expression of interleukin-6 (IL-6) and interleukin-10 (IL-10) in the lung of acute lung injury model mice. Methods Thirty Kunming mice were randomly divided into normal control group, acute lung injury group and Wortmannin treatment group. The average optical density of IL-6 mRNA in the lung tissue of Wortmannin-treated mice was (0.37 ± 0.03), which was significantly lower than that in the lung tissue of mice with acute lung injury -6 mRNA (0.26 ± 0.02), P <0.05; RT-PCR method to detect IL-10 mRNA in three groups of mice found that Wortmannin treated mice lung IL-10 mRNA average optical density (0.27 ± 0.02), which was significantly higher than that of the lung tissue of mice with acute lung injury (0.18 ± 0.02), P <0.05. Conclusion Wortmannin can inhibit the expression of IL-6 in lung tissue and up-regulate the expression of IL-10 in mice with acute lung injury.
