本文旨在观察血液灌流对健康大白鼠门静脉滴注5-Fu时血浆及组织浓度分布、外周血象的变化。在体外实验证实活性炭类吸附剂对5-Fu吸附效果极好。大白鼠门静脉滴注5-Fu(100mg/kg)30min后,血浆浓度在血液灌流组(DHP组)于30min及120min均分别明显低于对照组(P<0.05)。肝组织浓度则在两组间无显著差异,而心、肠、肾组织的浓度却分别低于对照组(P<0.05)。外周血象白细胞于给药后1,3天在玻珠对照灌流组中下降更明显(P<0.05)。而血小板下降程度则于对照组第3天更明显(P<0.05)。本实验结果提示,采用CAAC-Ⅱ血液灌流与门静脉滴注5-Fu联用,有助于减低外周血及肝外组织浓度,而保持肝组织中的药物含量。
This article aims to observe the changes of plasma and tissue concentration distribution and peripheral blood changes when hemoperfusion is administered to portal vein of healthy rats by 5-Fu infusion. In vitro experiments confirmed that activated carbon adsorbent adsorption of 5-Fu effect is excellent. The plasma concentration of 5-Fu (100mg / kg) 30 min after portal vein infusion in rats was significantly lower than that of the control group (P <0.05) at 30 min and 120 min in hemoperfusion group (DHP group). There was no significant difference in liver tissue concentration between the two groups, but the concentrations of heart, intestine and kidney tissue were lower than those of the control group (P <0.05). Peripheral blood leukocytes decreased more significantly in the bead-control perfusion group at day 1,3 after administration (P <0.05). The degree of thrombocytopenia in the control group on the 3rd day was more significant (P <0.05). The results suggest that the use of CAAC-Ⅱ hemoperfusion and portal vein infusion of 5-Fu combined to help reduce the peripheral blood and extrahepatic tissue concentrations, while maintaining the liver in the drug content.
