目的:探讨西地那非对糖尿病勃起功能障碍(ED)大鼠勃起功能的保护作用。方法:SD雄性成年大鼠55只,45只腹腔内注射链脲佐菌素(STZ)诱发糖尿病,10只注射生理盐水作对照。将糖尿病大鼠分为3组,其中一组喂食单纯淀粉颗粒,另两组为含西地那非淀粉颗粒,剂量分别相当于50kg体重20mg/d和50mg/d。6周后,大鼠颈部皮下注射阿朴吗啡(APO),录像记录大鼠阴茎勃起状况。结果:STZ诱发大鼠糖尿病成功率为86.7%。3组糖尿病大鼠6周存活率为84.6%、91.7%和85.7%。服用西地那非并不影响糖尿病大鼠生存率、血糖水平及体重。注射APO后,对照组勃起率为100.0%,糖尿病组仅为44.1%。糖尿病大鼠未服用西地那非、服用西地那非20mg/d或50mg/d各组勃起率和平均勃起次数分别为27.3%、36.4%、66.7%和0.36±0.2、0.54±0.3、1.10±0.6。比较平均勃起次数,前两组差异无显著性(P>0.05),但后组明显高于前两组,差异有显著性(P<0.05)。结论:持续服用西地那非有助于对糖尿病ED大鼠勃起功能的保护;增加西地那非剂量保护作用更为明显。
Objective: To investigate the protective effect of sildenafil on erectile function in diabetic erectile dysfunction (ED) rats. Methods: Fifty-five SD male adult rats were injected intraperitoneally with streptozotocin (STZ) to induce diabetes mellitus, and 10 rats were injected with normal saline. The diabetic rats were divided into three groups, one group fed simple starch granules, the other two groups containing sildenafil starch granules, the dosage was equivalent to 50kg body weight 20mg / d and 50mg / d. After 6 weeks, apomorphine (APO) was subcutaneously injected into the neck of the rats, and the penile erection of the rats was recorded by video. Results: The success rate of STZ-induced diabetic rats was 86.7%. The 6-week survival rates of three groups of diabetic rats were 84.6%, 91.7% and 85.7%. Sildenafil does not affect the survival rate of diabetic rats, blood glucose levels and body weight. After APO injection, the erection rate was 100.0% in the control group and 44.1% in the diabetic group. Diabetic rats did not take sildenafil, taking sildenafil 20mg / d or 50mg / d each group erection rate and average erection times were 27.3%, 36.4%, 66.7% and 0.36 ± 0.2,0.54 ± 0.3,1.10 ± 0.6. CONCLUSIONS: Sustained use of sildenafil helps to protect erectile function in diabetic ED rats and increases the dose-dependent protective effects of sildenafil.
