从769条含SSR位点的高粱EST序列中设计了103对EST-SSR引物。用设计出的103对EST-SSR引物,对高粱品种TX623A和苏丹草品种S722进行了PCR扩增和多态性检测。结果表明:80对引物同时在两品种中扩出了条带,占引物总数的77.7%,20对引物在两品种中表现出多态性,占引物总数的19.4%,占能在两品种中扩出带引物的25.0%。研究结果表明根据高粱EST建立高粱与苏丹草EST-SSR标记是有效、可行的。
103 pairs of EST-SSR primers were designed from 769 EST sequences of sorghum containing SSR loci. 103 pairs of EST-SSR primers were designed and used to detect the polymorphism of sorghum variety TX623A and sudan variety S722. The results showed that 80 pairs of primers amplified two bands simultaneously, accounting for 77.7% of the total number of primers, and 20 pairs of primers showed polymorphism in the two varieties, accounting for 19.4% of the total number of primers, Expand 25.0% of primer with primer. The results showed that it is effective and feasible to establish EST-SSR markers of sorghum and sorghum based on EST.
