目的:观察宫颈癌细胞HeLa转染chk2反义寡核苷酸后,在X射线作用下对凋亡的影响。方法:以脂质体为载体将chk2正义链和反义链转染HeLa细胞,用Western blot检测HeLa细胞中Chk2蛋白的表达,用流式细胞仪和TUNEL法检测X射线照射后细胞凋亡率。结果:反义封闭chk2可明显抑制Chk2蛋白表达(P<0.01),X射线作用下细胞凋亡率明显高于转染正义链组(P<0.01)。结论:反义封闭chk2基因可增加He-La细胞对X射线照射的凋亡敏感性。
OBJECTIVE: To observe the effect of HeLa on the apoptosis of human cervical cancer cells after transfection with chk2 antisense oligonucleotide. Methods: The chk2 sense and antisense strands were transfected into HeLa cells by lipofectamine. The expression of Chk2 protein in HeLa cells was detected by Western blot. The apoptosis rate of HeLa cells was detected by flow cytometry and TUNEL . Results: Antisense blocking of chk2 significantly inhibited the expression of Chk2 protein (P <0.01). The apoptotic rate of X-ray irradiation was significantly higher than that of transfected sense chain (P <0.01). Conclusion: Antisense chk2 gene can increase the sensitivity of He-La cells to X-ray irradiation.
