目的:建立高效液相色谱同时测定不同产地、不同品种绞股蓝药材中芦丁和槲皮素含量的方法。方法:ZORBAX SB-C18色谱柱(4.6 mm×150 mm,5μm),流动相乙腈(A)-0.2%磷酸溶液(B)梯度洗脱,流速1.0 m L·min-1,柱温30℃,检测波长360 nm,进样量10μL。结果:芦丁在2.535~10.14μg线性良好(R~2=0.999 2),平均回收率为98.72%,RSD 1.2%;槲皮素在0.018 1~0.144 8μg线性良好(R~2=0.999 3),平均回收率98.40%,RSD 1.7%。结论:该方法简便可行、重复性好,适用于绞股蓝药材2种有效成分的同时测定,可为绞股蓝药材的质量控制提供参考依据。
OBJECTIVE: To establish a method for simultaneous determination of rutin and quercetin in different genus Gynostemma by HPLC with simultaneous determination of different origins. METHODS: The mobile phase was eluted with a mobile phase of acetonitrile (A) - 0.2% phosphoric acid (B) at a flow rate of 1.0 m L · min-1 on a ZORBAX SB-C18 column (4.6 mm × 150 mm, Detection wavelength of 360 nm, injection volume 10μL. Results: The average recoveries of rutin from 2.535 to 10.14μg were good (R ~ 2 = 0.999 2), with an average recovery of 98.72% and a RSD of 1.2%. Quercetin was linear at 0.018 1 ~ 0.144 8μg (R ~ 2 = 0.999 3) , The average recovery rate of 98.40%, RSD 1.7%. Conclusion: The method is simple, feasible and reproducible. It is suitable for the simultaneous determination of two active ingredients in Gynostemma. It can provide a reference for the quality control of Gynostemma.
