目的观察加味七方胃痛颗粒对CAG模型大鼠bcl-2、cyclinD1蛋白表达的影响。方法将120只Wistar大鼠随机分为6组,分别为空白组、模型组、胃复春组、加味七方胃痛颗粒大、中、小剂量组,每组各20只。除空白组外,其余5组进行造模。造模成功后,模型组予生理盐水,胃复春组予胃复春片水溶液,加味七方胃痛颗粒组予加味七方胃痛免煎中药颗粒,1次/d。连续治疗30 d后,光镜下观察胃黏膜组织学改变及运用免疫组化的方法观察各大鼠bcl-2及cyclinD1蛋白的表达。结果与空白组相比,模型组大鼠bcl-2、cyclinD1蛋白阳性表达率显著升高(P<0.05);bcl-2、cyclinD1蛋白的阳性表达率加味七方胃痛颗粒组较模型组、胃复春组降低(P<0.05)。结论加味七方胃痛颗粒各治疗组较模型组胃黏膜的阳性表达率明显降低,表明加味七方胃痛颗粒能抑制bcl-2、cyclinD1蛋白的表达,调节细胞的凋亡和增殖,从而改善慢性萎缩性胃炎的胃黏膜病变。
Objective To observe the effect of Jiawei Qi Fang Wei Tong Granule on the expression of bcl-2 and cyclinD1 protein in CAG model rats. Methods 120 Wistar rats were randomly divided into 6 groups: blank group, model group, Weifuchun group, Jiawei Qifangwei Tong Granule large, medium and small dose groups of 20 each. In addition to the blank group, the remaining five groups for modeling. After the model was established successfully, the model group was treated with normal saline and Weifuchun with the aqueous solution of Weifuchun tablets. The modified Qifangweitong granule group was treated with modified Qifangweihuagong decoction granules once a day. After 30 days of continuous treatment, histological changes of gastric mucosa were observed under light microscope and the expression of bcl-2 and cyclinD1 protein in each rat was observed by immunohistochemistry. Results Compared with the blank group, the positive expression rates of bcl-2 and cyclinD1 in the model group were significantly increased (P <0.05). The positive expression rate of bcl-2 and cyclinD1 in the model group was significantly higher than that in the model group and the stomach Rehabilitation group decreased (P <0.05). Conclusions The positive expression rate of gastric mucosa in Jiawei Qifangwei Tongtong Granules group is lower than that in model group, indicating that Modified QiFangWeiTong Granules can inhibit the expression of bcl-2 and cyclinD1 protein, regulate the apoptosis and proliferation of cells, and improve the chronic atrophy Gastritis Gastric mucosal lesions.
