PF1 8 3抗体是一株大鼠抗小鼠胸腺基质细胞单克隆抗体 .在以往的共培养研究中发现该抗体可抑制小鼠胸腺树突状细胞系 (MTSC4)诱导的胸腺细胞凋亡作用 .目的是观察PF1 8 3抗体识别分子 (PF1 8 3分子 )的特点及其在MTSC4诱导的胸腺细胞凋亡中的作用 .利用流式细胞仪对PF1 8 3分子的表达进行了特性分析 ,发现PF1 8 3分子除表达于MTSC4表面外 ,还表达于ConA活化的胸腺细胞上 ,但新鲜胸腺细胞表面无表达 .胸腺细胞与MTSC4共育 48h后 ,PF1 8 3分子表达上调 .流式细胞仪分析PI染色检测凋亡相关亚二倍体及PF1 8 3抗体染色的结果表明 ,PF1 8 3分子特异表达于高亚二倍体含量的凋亡胸腺细胞 .经免疫共沉淀和Westernblotting鉴定 ,凋亡胸腺细胞表达的PF1 8 3分子的分子量为 35ku .该分子可能参与胸腺细胞凋亡过程 .
PF1 8 3 antibody is a rat anti-mouse thymic stromal cell monoclonal antibody that was previously found to inhibit the thymocyte apoptosis induced by mouse thymic dendritic cell line (MTSC4) in a co-culture study.
