目的:探讨人4IgB7-H3基因转染人胃癌细胞株SGC7901后人类β1,3半乳糖基转移酶7(β1,3-galacto-syltransferase 7,β3GalT7)的表达变化,以期发现4IgB7-H3与β3GalT7的相互关系。方法:脂质体介导重组逆转录病毒载体pEGZ-Term/4IgB7-H3转染SGC7901细胞,筛选获得稳定表达4IgB7-H3的亚克隆细胞株,通过RT-PCR和蛋白质印迹方法检测β3GalT7的表达。结果:成功获得稳定表达4IgB7-H3的亚克隆细胞株SGC7901/4IgB7-H3;β3GalT7随4IgB7-H3的表达上调而上调。结论:4IgB7-H3蛋白的修饰成熟可能需要β3GalT7的作用。
OBJECTIVE: To investigate the expression of human β1,3-galacto-syltransferase 7 (β3GalT7) after human 4IgB7-H3 gene transfection in human gastric cancer cell line SGC7901 in order to find out the relationship between 4IgB7-H3 and β3GalT7 Interrelationship. METHODS: SGC7901 cells were transfected with liposome-mediated recombinant retroviral vector pEGZ-Term / 4IgB7-H3, and subclone cell lines stably expressing 4IgB7-H3 were screened. The expression of β3GalT7 was detected by RT-PCR and Western blotting. Results: The subclone cell line SGC7901 / 4IgB7-H3 stably expressing 4IgB7-H3 was successfully obtained and β3GalT7 was up-regulated with the increase of 4IgB7-H3. Conclusion: The modification of 4IgB7-H3 protein may require the effect of β3GalT7.
